Mechanisms of Microglia Proliferation in a Rat Model of Facial Nerve Anatomy

Although microglia exist as a minor glial cell type in the normal state of brain, they increase number response to various disorders and insults. However, it remains unclear whether proliferate affected area, mechanism proliferation has long attracted attention researchers. We analyzed microglial mitosis using facial nerve transection model which blood–brain barrier is left unimpaired when nerves are axotomized. Our results showed that levels macrophage colony-stimulating factor (M-CSF), cFms (the receptor for M-CSF), cyclin A/D, proliferating nuclear antigen (PCNA) were increased axotomized nucleus (axotFN). In vitro experiments revealed M-CSF induced cFms, PCNA microglia, suggesting vivo. addition, caused activation c-Jun N-terminal kinase (JNK) p38, specific inhibitors JNK p38 arrested mitosis. shown play roles induction cyclins/PCNA respectively. was suggested be through signaling cascade p38-mitogen- stress-activated kinase-1 (MSK1)-cAMP-responsive element binding protein (CREB) and/or p38-activating transcription 2 (ATF2). Microglia axotFN anticipated serve neuroprotective cells by supplying neurotrophic factors scavenging excite toxins reactive oxygen radicals.
